How This Calculator Works

The Michaelis-Menten equation describes how the rate of an enzyme reaction depends on substrate concentration. At low substrate levels the rate rises almost linearly, and as substrate increases the rate approaches a maximum velocity called Vmax. The Michaelis constant Km is the substrate concentration that produces half of Vmax, and it reflects how tightly the enzyme binds its substrate.

The reaction velocity equals Vmax times the substrate concentration divided by the sum of Km and the substrate concentration. The calculator also reports what fraction of the maximum velocity the enzyme is achieving.

Clinical and Industrial Relevance

Michaelis-Menten kinetics underpin drug dosing, toxicology, and bioprocess design. Ethanol metabolism in the liver follows saturation kinetics: at low concentrations the rate is roughly first-order, but above ~100 mg/dL the enzyme (alcohol dehydrogenase) is nearly saturated and metabolism proceeds at a constant rate (~15 mg/dL/hour in most adults). This is why blood alcohol clears at a roughly fixed hourly rate regardless of how much was consumed - a direct consequence of Vmax-limited kinetics.

In pharmacokinetics, drugs with Km well above therapeutic plasma concentrations show linear (first-order) clearance; those with Km near therapeutic concentrations show nonlinear kinetics - small dose increases produce disproportionate plasma level rises. Phenytoin is the classic example: small dose adjustments can push a patient from sub-therapeutic to toxic levels. In industrial bioprocessing, knowing Km and Vmax allows engineers to design bioreactors at the optimal substrate concentration that maximizes volumetric productivity without wasting substrate.
